I have an ODB2 diagnostic reader for my W203 C180 but recently I can't connect anymore and it doesn't give any output.
How do I troubleshoot if there is a problem with the ODB2 interface?

Perhaps you would like to describe what OBD2 tool you are using, maybe include a photo of the interface?
 
Do you have another car you could try or another diagnostic tool? What specific tool are you using that you are having problems with?
 
Check the power and ground lines from the OBD connector, the fuse may have blown and that's why you can't connect to the ECU.

Sometimes the problem is broken contacts, but most often it is software related or software and OBD are not compatible, sometimes OBD2 is sold that is only compatible with Android, etc. If the problem is in the app, I can recommend a popular app called Car Scanner in the App Store.
 
For those who are interested. When using the OBD-USB adapter, you need to correctly configure the port (COM1 - COMX) in Windows and in the configuration of the software used. I also had problems with the baud rate, but this is for a very specific tool.
 
Why connect with a wire, try connecting via Bluetooth? I checked codes this way on my previous car.
 
Check 12V power (pin 16) and ground (pin 4, 5) at the OBD connector.
